This paper presents an analysis of pteridophyte spore bank the site of Pedregulho, in Itirapina, state of São Paulo, Brazil. The samples of soil were collected in February (rainy season) and August (dry season) in four localities: open cerrado, cerradão, gallery forest and a disturbed area at depths of 0-5, 5-10 and 10-15 cm. The samples were collected in duplicate. Of the duplicates, one was used for spore counting and the second one for spore germination. In the cerrado, cerradão and gallery forest soil samples the values for pteridophyte spores were similar in the rainy and dry season, but in the disturbed area the percentage of spores was higher in the rainy season than in the dry season. The germination of three cerrado herbaceous species: Bidens gardneri, Vernonia herbacea (Asteraceae) and Psychotria barbiflora (Rubiaceae) was studied using seeds stored in cerrado soil and in refrigerator at 4°C. The field experiment was carried out at a preserved area of cerrado, the Reserva Biológica e Estação Experimental de Mogi-Guaçu, State of São Paulo, Brazil. The soil was collected in the same cerrado where the experiment was carried out. The achenes of Bidens gardneri are light sensitive, but this sensitivity disappeared when the achenes were stored in soil. The spores used were collected from a population of trees of Cyathea delgadii Sternb. growing in a gallery forest at the Reserva Biológica e Estação Experimental de Moji Guaçu, São Paulo state, Brazil (22°18’S and 47°11’W). The germination of spores of Cyathea delgadii decreases with time when kept in closed bottles under storage at 4°C in darkness. Germination is still very high after storage for one year. Spores stored for three years do not germinate. The results also show a decrease in soluble proteins and an increase in starch after several months storage.
